ABSTRACT

Objective:To investigate the effect of high-intensity interval training on cognitive weakness in elderly patients with coronary heart disease. Methods:A randomized controlled study was used. A total of 70 elderly patients with coronary heart disease after discharge from the Cardiology Department, Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine from June 2020 to June 2021 were selected and divided into the control group (35 cases) and the observation group (35 cases) by random digits table method. The control group received conventional treatment, and the observation group received high-intensity interval training on the basis of conventional treatment. The changes of peak oxygen intake (PeakVO 2), anaerobic threshold oxygen uptake (ATVO 2), Fried phenotype score, Montreal Cognitive Assessment score, and SF-36 quality of life assessment score before and after 12 weeks of intervention were compared between the two groups. Results:During the intervention, 2 cases fell off in the control group and 2 cases in the observation group. After 12 weeks of intervention, PeakVO 2 and ATVO 2 in the observation group were (23.91 ± 5.88), (20.79 ± 5.19) ml·min -1·kg -1, respectively, higher than those in the control group(20.56 ± 5.81), (17.29 ± 5.36) ml·min -1·kg -1, and the differences were statistically significant ( t = 2.38, 2.69, both P<0.05). The scores of SF-36 quality of life assessment and Montreal Cognitive Assessment in the observation group were (85.33 ± 6.43), (22.64 ± 3.81) points, which were significantly higher than those in the control group (78.72 ± 8.14), (20.67 ± 3.04) points, and the differences were statistically significant ( t = 3.66, 2.32, both P<0.05); the Fried phenotype score in the observation group was (1.36 ± 0.99) points, which was significantly lower than that in the control group (3.03 ± 1.08) points, and the difference was statistically significant ( t = 6.54, P<0.05). Conclusions:High-intensity interval training can improve cardiopulmonary function, aerobic exercise ability, cognitive weakness and quality of life in elderly patients with coronary heart disease.

ABSTRACT

Objective An optimal radiation dose was made research on tumor cells to develop effective tumor fusion vaccines. Methods We used the RS 2000 biological X-ray radiation instrument to produce different dose of X-rays.Mouse liver cancer cells line,BNL 1ME a.7R.1(MEAR),was used for experiment. The RS 2000 biological X-ray radiation was applied to create different tumor cell clones, which could help us to study the rela-tionship between irradiation dose and tumor cells′proliferation activity.Furthermore, the fusion cells′anti-tumor ef-fect was examined by flow cytometry. Results High-dose radiation would induce the lower proliferation of cancer cells than low-dose irradiation do.Conclusions During the preparation of fusion vaccines, irradiation dose should be considered as a factor that would influence the tumor cells′ proliferation activity. When the dose of irradiation was appropriate,we could make safe and efficient integration cell vaccines.

ABSTRACT

The objective of the study was to develop a simple, specific and stability-indicating HPLC method for the simultaneous determination of creatine phosphate sodium [CPS] and its related substances in pharmaceutical formulation. Separation of creatine phosphate sodium from its major process impurities and degradation products was achieved on a Hypersil BDS C18 column [250 × 4.6 mm, 5 microm] with an aqueous mobile phase containing 0.2% [w/v] tetrabutylammonium hydroxide [TAH] and 0.2% [w/v] monopotassium phosphate adjusted to pH 6.6 with orthophosphoric acid at a flow rate of 1.0 mL min-1. The analytes were detected at 210 nm. Different chromatographic parameters were carefully optimized. The relative response factors for creatine, creatinine and creatinine phosphate disodium salt relative to CPS were determined. The method has been validated with respect to solution stability, system suitability, LOD, LOQ, linearity, accuracy, precision, specificity and robustness. The validation criteria were met in all cases. The developed method was successfully applied to determine the purity of CPS in pharmaceutical formulation

ABSTRACT

Objective To observe the effect of moxibustion on the blood lipid and hemorheology in senile patients with coronary heart disease (CHD) due to heart blood stagnation who underwent interventional treatment.Method A hundred senile CHD patients were randomized into a treatment group and a control group, 50 cases in each group. The control group was intervened by conventional medications, while the treatment group was additionally by thunder-fire moxibustion. The blood lipid and hemorheology were observed before the treatment and after 3 treatment courses.Result The blood lipid parameters (including CHO, TG, HDL-C and LDL-C) were changed significantly in the treatment group after the intervention (P<0.05). The CHO, TG, and LDL-C levels of the treatment group were significantly different from that of the control group (P<0.05). The hemorheology parameters (whole blood viscosities at a high-, middle-, and low-shear rate, and fibrinogen) were significantly changed in the treatment group after the intervention (P<0.05). The whole blood viscosities at a high-, middle-, and low-shear of the treatment group were markedly different from that of the control group (P<0.05).Conclusion Thunder-fire moxibustion can down-regulate the blood lipid and blood viscosity of CHD patients undergone interventional treatment, and thus improve myocardial ischemia.

ABSTRACT

Objective To analyze the change of cysteinyl leukotriene ( Cys-LTs) levels and 8-Isopros-tane (8-iso-PG) levels in the exhaled breath condensate (EBC) of asthmatic children from acute exacerbation to clinical remission, and investigate the role of the detection of Cys-LTs and 8-iso-PG in EBC in its severity and pathogenesis , and explore the relationship between the Cys-LTs and 8-iso-PG through measuring Cys-LTs levels and 8-iso-PG levels in the EBC of asthmatic children. Methods The outpatient or inpatient asthmatic children of the pediatrics and a group of healthy children were studied. All subjects′ EBC were collected by the R-Tube produced by American Respiratory Research. The concentration of Cys-LTs and 8-iso-PG in EBC were measured by enzyme-linked immunosorbent assay (ELISA), and compared among children in asthmatic exacerbation, asthmatic remission, and healthy condition. The relevance of their change would be explored at the same time. Results (1) Cys-LTs levels in EBC were higher in asthma exacerbation, compared to healthy controls (P0. 05 ) . ( 2 ) 8-iso-PG levels was higher in asthmatic exacerbation compared to asth-matic remission ( P<0. 05 ) . Moreover, the 8-iso-PG levels were significantly higher in asthmatic remission than in healthy controls (P<0. 05). (3) Through the relevance analysis of the Cys-LTs and 8-iso-PG levels in EBC among the three groups, Cys-LTs levels in EBC of asthmatic exacerbation significantly were correlated with 8-iso-PG levels (n1 =35, r1 =0. 61, P<0. 05), while there was no significant correlation between 8-iso-PG levels and Cys-LTs levels in asthmatic remission. Conclusion The increase of 8-iso-PG levels in EBC of bronchial asthmatic patients correlates with the disease and its control. Therefore, 8-iso-PG can be an objective indicator for asthmatic diagnosis and healing efficacy. Cy-LTs levels increase in the EBC of bronchial asthmatic according to disease severity. The two levels correlate during asthmatic exacerbation, indicating that a link be-tween airway oxidative stress and inflammation among asthmatics.

ABSTRACT

RNA interference (RNAi) has exhibited huge potentials on anti-HIV-1 therapy research. The obtainment of RNAi element targeting to HIV-1 highly effectively and specifically was crucial for relevant research. Recent reports had described that microRNAs (miRNAs) posses more characteristics of inhibition and expression mechanisms than small interfering RNAs (siRNAs). In this study we explored the construction of artificial miRNA targeting to HIV-1 effectively and specifically. Sixteen siRNAs sequences were selected based on the conserved regions in the HIV-1 pol gene. ShRNA expression vectors were co-transfected with HIV-1 clone pNL4-3 to evaluate the abilities of siRNAs to inhibit HIV-1 expression. The pol1026 sequence was selected from candidates. The target sequence in the stem-loop structure of the well-characterized native miR-30a was replaced with pol1026 sequences, and the artificial miRNA expression vectors were co-transfected with the HIV-1 clone pNL4-3, results showed that HIV-1 can be effectively inhibited by miR-1026E. Target specificity of miR-1026E was confirmed by co-transfection assay with reporter plasmids containing different target sequences. The miR-1026E expression element was then inserted into Lentivirus which was used as a vector to transduce the MT-4 cells, MT-4-miR1026E expressing miR-1026E stably was cloned from transduced cells. The MT-4-miR1026E cell effectively inhibited HIV-1 replication in vitro. And the intracellular miR-181 and miR-16 expression levels and statl mRNA levels were not affected by the expression of miR-1026E in MT-4-miR1026E cells. miR-1026E is a promising candidate for future research.

ABSTRACT

Objective To explore the biological effects of the luteinizing hormone receptor (LHR) and vascular endothelial growth factor (VEGF) on the ovary of mice during peri-implantation. Methods The immunohistochemistry SABC method and image analysis were used to study the distribution and changes of the LHR and VEGF in Kunming mouse( n =28) ovary during estrous, pregnancy of day 1, day 4 and day 6 stage. Results The expression of LHR-immunoreactive substance and that of VEGF-immunoreactive substance had the same distribution and changes. Compared with other groups, the level of LHR-immunoreactive substance and that of VEGF-immunoreactive substance increased highly on the stroma cells around largergrowing follicles in estrous group ( P <0.05). Along with the pregnancy, the positive immunostaining for LHR and VEGF increased gradually on the granulosa lutein cells, and reached the highest level on day 6 of pregnancy. Positive immunostaining for LHR or VEGF on some endothelia and blood cells were observed in day 1 of pregnancy or estrous group respectively. Form day 1 of pregnancy, the theca cells had positive immunostaining for LHR. Conclusion The expression of LHR and VEGF is closely related with the process of follicle growing, ovulation and corpus luteum formation.
